运用代谢组学分析方法评价当归不同炮制品挥发油抗大鼠急性炎症的作用及其机制。采用当归炭挥发油(C-VOAS)、酒当归挥发油(J-VOAS)、土当归挥发油(T-VOAS)和油当归挥发油(Y-VOAS)分别干预角叉菜胶诱导大鼠足肿胀急性炎症模型,检测大鼠血浆HIS,5-HT,PGE2和TNF-α含量的变化。并应用气相色谱-质谱联用技术检测各组大鼠血浆代谢指纹图谱,应用主成分分析和偏最小二乘判别分析和单变量统计分析方法探讨血浆中与炎症相关差异代谢物的变化。结果显示C-VOAS和J-VOAS对炎性介质HIS,5-HT,PGE2和细胞因子TNF-α具有极显著的抑制效果(P<0.01),T-VOAS和Y-VOAS对其亦具有显著的抑制效果(P<0.05)。代谢组学分析发现,与正常对照相比,大鼠急性炎症模型血浆中14个内源性差异代谢物的代谢被干扰(P<0.05或P<0.01);与急性炎症模型组相比,J-VOAS和C-VOAS能有效地回调多数代谢物水平,而T-VOAS和Y-VOAS的回调能力次之(P<0.05或P<0.01)。研究表明当归的不同炮制品挥发油对角叉菜胶致大鼠足肿胀急性炎症均有不同程度的抑制作用,作用途径各有特点,其中J-VOAS和C-VOAS抗炎效果最好,Y-VOAS和T-VOAS次之。因此,代谢组学研究方法可应用于阐明当归不同炮制品挥发油抗炎作用机制,为当归不同炮制品的临床应用提供了理论基础。
Metabolomics analysis was used to evaluate the effect and mechanism of volatile oil from Angelica sinensis Angelica on acute inflammation in rats. C-VOAS, J-VOAS, T-VOAS and Y-VOAS were used to induce carrageenan-induced acute inflammation model in rats The changes of plasma HIS, 5-HT, PGE2 and TNF-α in rats were detected. The plasma metabolic fingerprints of rats in each group were detected by gas chromatography-mass spectrometry. Principal component analysis, partial least-squares discriminant analysis and univariate statistical analysis were used to investigate the changes of plasma metabolites related to inflammation. The results showed that C-VOAS and J-VOAS had a significant inhibitory effect on inflammatory mediators HIS, 5-HT, PGE2 and cytokine TNF-α (P <0.01). T-VOAS and Y-VOAS also had significant inhibitory effect (P <0.05). Metabolomics analysis showed that the metabolism of 14 endogenous differentially expressed metabolites in the acute inflammation model of rats was disturbed compared with the normal control (P <0.05 or P <0.01). Compared with the acute inflammation model group, J -VOAS and C-VOAS can effectively call back most of the metabolites, while T-VOAS and Y-VOAS have the second lowest callback ability (P <0.05 or P <0.01). The results showed that different processed products of Angelica Angelica volatile oil carrageenan induced acute foot inflammation in rats with varying degrees of inhibition, the role of the pathways have their own characteristics, including J-VOAS and C-VOAS anti-inflammatory effect of the best, Y- VOAS and T-VOAS second. Therefore, the metabonomics research method can be applied to clarify the anti-inflammatory mechanism of the volatile oil of Angelica sinensis different processed products, which provides a theoretical basis for the clinical application of different processed products of Angelica sinensis.
