目的探讨联合检测外周血端粒酶活性和组织多肽特异性抗原对乳腺癌患者的临床价值。方法采用聚合酶链反应(PCR)-端粒酶重复序列扩增反应(TRAP)-酶联免疫吸附测定(ELISA)法,分别检测67例乳腺癌患者、31例乳腺良性病变患者及36例体检健康者的外周血单个核细胞的端粒酶活性;同时对其血清应用ELISA法检测组织多肽特异性抗原的表达情况。结果外周血端粒酶活性和组织多肽特异性抗原的表达均与乳腺癌淋巴结转移密切相关(P<0.05)。其中,外周血端粒酶活性的表达还与癌组织的浸润相关(P<0.001)。结论外周血端粒酶活性和组织多肽特异性抗原的表达预示乳腺癌淋巴结转移,并且外周血端粒酶活性的表达还与乳腺癌的局部浸润生长有关。联合检测这2项指标,对患者预后的评估及治疗方案的制定具有重要的意义。
Objective To investigate the clinical value of combined detection of telomerase activity and tissue polypeptide-specific antigen in peripheral blood of patients with breast cancer. Methods Totally 67 cases of breast cancer, 31 cases of benign breast lesions and 36 cases of physical examination were detected by polymerase chain reaction-polymerase chain reaction-restriction fragment length polymorphism (TRAP) and enzyme-linked immunosorbent assay (ELISA) Healthy people’s peripheral blood mononuclear cells of telomerase activity; At the same time, its serum ELISA method was used to detect the expression of tissue polypeptide-specific antigen. Results The expression of telomerase activity and tissue polypeptide specific antigen in peripheral blood were closely related to lymph node metastasis of breast cancer (P <0.05). Among them, the expression of telomerase activity in peripheral blood was also related to the infiltration of cancer tissues (P <0.001). Conclusion The expression of telomerase activity and tissue polypeptide-specific antigen in peripheral blood indicates the lymph node metastasis in breast cancer, and the expression of telomerase activity in peripheral blood is also related to the local infiltration growth of breast cancer. Joint detection of these two indicators, the prognosis of patients with treatment and the development of programs of great significance.
