硫氰钴铵比色法检测流感裂解疫苗中Triton N-101残留量

目的用硫氰钴铵比色法检测流感裂解疫苗中Triton N-101残留量。方法用硫氰钴铵显色,620nm测定吸光度,检测Triton N-101的线性、空白回收率、加标回收率和精密度。结果 Triton N-101含量在0~400μg/ml时,线性良好,R2大于0.99;空白回收率在87.4%~95.4%之间,加标回收率在94.7%~99.8%之间;精密度(CV%)≤6%。结论该方法能满足流感裂解疫苗中Triton N-101残留量检测。 Objective To determine the residual amount of Triton N-101 in influenza lysed vaccine by ammonium thiocyanate cobalt colorimetry. Methods The color of thiocobalium chromite was determined by measuring the absorbance at 620 nm. The linearity and blank recovery of Triton N-101 were determined, and the recoveries and precision of spiked Triton N-101 were detected. Results The linearity of Triton N-101 ranged from 0 to 400 μg / ml with R2 greater than 0.99. The recoveries ranged from 87.4% to 95.4% with recoveries of 94.7% -99.8%. The precision (CV %) ≤6%. Conclusion This method can meet the detection of Triton N-101 residue in influenza lytic vaccine.
