目的分离家族性急性髓系白血病患者骨髓中差异表达的基因,在基因水平上探讨急性白血病发生发展的分子机制。方法应用 Super SMART 和抑制性消减杂交(SSH)技术,构建家族性急性髓系白血病 cDNA 消减文库;应用差异筛选技术进行差异表达片段的筛选与鉴定;测序结果进行 BLAST 同源性比对分析。结果成功构建了家族性急性髓系白血病 cDNA 消减文库;克隆、筛选并鉴定了17条基因片段,与已知基因同源,11条为未知基因片段。结论 SSH 技术是筛选差异表达基因的有效方法;获得多个与细胞增殖和分化相关的已知基因;获得11条新基因 EST 片段,并被GenBank 收录。
Objective To isolate the genes differentially expressed in the bone marrow of patients with familial acute myeloid leukemia and to explore the molecular mechanism of the development of acute leukemia at the gene level. Methods Super SMART and SSH were used to construct cDNA subtractive library of familial acute myeloid leukemia. Differential screening was used to screen and identify differentially expressed fragments. Sequencing results were compared using BLAST homology analysis. Results A cDNA subtractive library of familial acute myelogenous leukemia was successfully constructed. Seventeen gene fragments were cloned, identified and identified, and 11 were unknown gene fragments. Conclusion SSH is an effective method to screen differentially expressed genes. Several known genes related to cell proliferation and differentiation were obtained. ESTs of 11 new genes were obtained and were reported by GenBank.
