目的研究稳心颗粒(WXKL)对血管紧张素Ⅱ(AngⅡ)诱导H9C2细胞肥大模型骨架蛋白的影响。方法体外培养H9 C2细胞,经血管紧张素Ⅱ诱导复制心肌细胞肥大模型,加入5 g/L稳心颗粒后,观察细胞的形态变化以及对骨架蛋白的影响。结果经AngⅡ诱导的H9C2心肌细胞形态发生改变,细胞横向拉伸,肿胀变大,细胞体积较正常心肌细胞增大;加入稳心颗粒后H9 C2心肌细胞形态有所恢复,但还是比正常细胞横向宽度增加。应用激光共聚焦显微镜下观察细胞形态发现,正常H9 C2细胞经10-7mol/L AngⅡ刺激48 h后,细胞形态明显肿胀变大,细胞的长度和宽度分别由(120.32±8.36)μm和(51.82±8.62)μm增加到(182.67±7.81)μm和(92.22±7.56)μm。加入稳心颗粒作用24 h后,AngⅡ组的H9C2心肌细胞形态有所恢复,细胞的长度和宽度为(160.35±5.66)μm、(55.87±4.52)μm。结论稳心颗粒在AngⅡ诱导的肥厚心肌H9C2细胞模型中,能改善细胞骨架蛋白,起到一定的改善心肌肥厚的作用。
Objective To investigate the effects of WXKL on angiogenesis-induced H9C2 cytoskeleton hypertrophy model. Methods H9 C2 cells were cultured in vitro. Cardiomyocytes hypertrophy was induced by angiotensin Ⅱ. After adding 5 g / L Wenxin granule, the morphological changes of cells and the effect on skeletal protein were observed. Results The H9C2 cardiomyocytes induced by AngⅡwere changed in morphology, the cells were stretched in the transverse direction, the swelling became larger and the volume of the cells increased than that of the normal cardiomyocytes. The shape of H9 C2 cardiomyocytes was restored after adding WXC, but still more than the normal cells Increased width. The morphological changes of the cells were observed under confocal laser scanning microscope (LSCM). The results showed that the cell morphology was obviously swollen and the length and width of cells were (120.32 ± 8.36) μm and (51.82 ± 8.62) μm to (182.67 ± 7.81) μm and (92.22 ± 7.56) μm. The morphology of H9C2 myocardial cells in AngⅡ group was recovered after 24 hours of treatment with Wenxin Granule. The length and width of H9C2 cells were (160.35 ± 5.66) μm and (55.87 ± 4.52) μm, respectively. Conclusion Wenxin granule can improve cytoskeletal protein in hypertrophic myocardium H9C2 induced by AngⅡ, and play a role in improving cardiac hypertrophy.
