目的筛选与涎腺腺样囊性癌转移相关的Notch信号通路基因。方法对已构建的涎腺腺样囊性癌高低转移细胞株ACC-M、ACC-2基因表达谱进行生物信息学分析,在与Notch信号通路基因进行比对后,筛选出差异基因,通过荧光实时定量PCR技术对相关基因的表达差异进行初步验证。结果涎腺腺样囊性癌的表达谱中有46个基因与Notch信号通路有关。验证发现,Notch-1,Notch-2,Notch-4,CHUK,FOXC1,FZD2,FZD3,GBP2和RUNX1在2个细胞株中的表达差别有统计学意义(P<0.05),同时Notch-1在发生转移的涎腺腺样囊性癌中的表达明显高于未发生转移的涎腺腺样囊性癌(P<0.05)。结论Notch信号通路中的多个基因可能参与了涎腺腺样囊性癌的发生和发展;Notch-1的表达与涎腺腺样囊性癌的发生、转移密切相关。
Objective To screen Notch signaling pathway genes involved in salivary adenoid cystic carcinoma metastasis. Methods The gene expression profiles of ACC-M and ACC-2 in human salivary adenoid cystic carcinoma cell lines were analyzed by bioinformatics methods. After comparison with Notch signaling pathway genes, the differentially expressed genes were screened by fluorescence Real-time quantitative PCR was used to verify the difference of the expression of related genes. Results There were 46 genes in the expression pattern of salivary adenoid cystic carcinoma related to the Notch signaling pathway. The results showed that the expression of Notch-1, Notch-2, Notch-4, CHUK, FOXC1, FZD2, FZD3, GBP2 and RUNX1 in two cell lines was significantly different (P <0.05) The metastasis of salivary adenoid cystic carcinoma was significantly higher than that without metastasis of salivary adenoid cystic carcinoma (P <0.05). Conclusions Multiple genes in Notch signaling pathway may be involved in the development and progression of salivary adenoid cystic carcinoma. The expression of Notch-1 is closely related to the occurrence and metastasis of salivary adenoid cystic carcinoma.
