目的 :研究电穿孔技术对小分子离子型药物经皮渗透的影响。方法 :应用双室扩散池方法研究电穿孔技术对萘普生在离体大鼠腹部皮肤经皮渗透的影响 ,并与被动扩散和离子导入进行比较。结果 :外加电脉冲 (指数衰减型脉冲 ,脉冲幅度为 4 0 0V ,电容器电容为 2 .2 μF ,脉冲率为 2 0pulses·min- 1,脉冲宽度τ≈ 6.0ms)或离子导入 ( 1mA·cm- 2 ,6h)时 ,萘普生的渗透速率和累积渗透量均大于被动扩散。外加脉冲的促透作用显著地低于离子导入。结论 :电穿孔技术能明显促进小分子离子型药物萘普生的经皮渗透。
Objective: To study the effect of electroporation on the transdermal permeation of small molecule ionic drugs. Methods: The effect of electroporation on the transdermal permeation of abdomen skin of naproxen rats was studied by double chamber diffusion cell method, and compared with passive diffusion and iontophoresis. Results: A series of pulses (exponential decay pulse, pulse width of 4.0V, capacitor capacitance of 2.2μF, pulse rate of 20pulses · min-1, pulse width of τ≈ 6.0ms) or iontophoresis - 2, 6h), the permeation rate and cumulative infiltration of naproxen were higher than that of passive diffusion. Impulsive permeation is significantly lower than iontophoresis. Conclusion: Electroporation can significantly promote the transdermal penetration of small molecule ionic drug naproxen.
