灵芝抗衰老活性多糖GLA的化学研究

灵芝子实体经热水提取,乙醇分级沉淀,除游离蛋白质和小分子杂质,得到具有免疫调节作用及抗氧自由基活性的多糖GLA。进一步分离纯化,得到8个多糖均一体,对其中主要成分GLA_4、GLA_7、GLSP_(1~3)5个多糖均一体进行了化学结构研究。以完全酸水解、过碘酸盐氧化、Smith降解、光谱测定及气相色谱分析等证明,GLA_4、GLA_7为以葡萄糖为主的中性杂多糖,GLSP_1为鼠李半乳聚糖肽,GLSP_2及GLSP_3为葡聚糖肽。它们的精甙键类型均以β-(1→3)(1→6)及(1→4)为主,GLA_4尚有α-甙键存在。 The Ganoderma lucidum fruiting body was extracted with hot water and ethanol was fractionated and precipitated. In addition to free protein and small molecule impurities, polysaccharide GLA with immunomodulatory activity and antioxidant activity was obtained. After further separation and purification, 8 polysaccharides were integrated, and the chemical structure of the five main polysaccharides, GLA_4, GLA_7, and GLSP_(1-3), was integrated. The complete acid hydrolysis, periodate oxidation, Smith degradation, spectrometry and gas chromatographic analysis demonstrated that GLA_4 and GLA_7 are glucose-based neutral heteropolysaccharides, GLSP_1 is rhamnogalactan peptides, GLSP_2 and GLSP_3 For glucan peptides. The type of fine-bored bonds are mainly β-(1→3) (1→6) and (1→4), and GLA_4 still has α-甙 bonds.
