目的:建立高效液相色谱法同时测定陈皮药材中5种黄酮类化合物柚皮芸香苷、橙皮苷、川陈皮素、3,5,6,7,8,3’,4’-七甲氧基黄酮及红橘素的含量,并对结果进行聚类分析。方法:采用Waters Symmetry C18色谱柱(250mm×4.6mm,5μm);流动相为0.1%甲酸水溶液-乙腈,梯度洗脱;检测波长为300nm;流速1.0mL.min-1。结果:柚皮芸香苷、橙皮苷、川陈皮素、3,5,6,7,8,3’,4’-七甲氧基黄酮及红橘素的线性范围分别为1.00~9.00μg.mL-1(r=0.9998)、1.00~9.00μg.mL-1(r=0.9999)、0.10~0.90μg.mL-1(r=0.9997)、0.10~0.90μg.mL-1(r=0.9999)、0.10~0.90μg.mL-1(r=0.9994);加样回收率(n=5)分别为96.8%,99.4%,93.3%,94.4%,95.8%。结论:该方法准确、简便,具有良好的重复性和稳定性。试验结果显示,全国不同省市、不同产地陈皮饮片的含量存在差异。同时也说明多指标成分定量分析比单一指标成分定量分析能更全面地反映陈皮饮片的内在质量。
Objective: To establish a HPLC method for the simultaneous determination of five flavonoids rutin, hesperidin, nobiletin, 3,5,6,7,8,3 ’, 4’-heptamethoxymethane Flavonoids and tangerine content, and cluster analysis of the results. Methods: A Waters Symmetry C18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of 0.1% formic acid in water and acetonitrile. The detection wavelength was 300 nm and the flow rate was 1.0 mL · min-1. Results: The linear range of ricin, hesperidin, nobiletin, 3,5,6,7,8,3 ’, 4’-heptamethoxyflavone and tangerin were 1.00 ~ 9.00μg. (r = 0.9997), 0.10-0.90 μg · mL-1 (r = 0.9999) , 0.10 ~ 0.90μg.mL-1 (r = 0.9994), respectively. The recoveries were 96.8%, 99.4%, 93.3%, 94.4% and 95.8%, respectively. Conclusion: This method is accurate, simple, and has good repeatability and stability. The test results show that there are differences in the content of Citrus Pieces from different provinces and cities and from different areas in China. At the same time, it also shows that the quantitative analysis of multiple index components can reflect the inner quality of dried tangerine peel slices more completely than the quantitative analysis of single index components.
