目的:探讨马钱子水提液对斑马鱼胚胎发育的影响,建立马钱子对斑马鱼胚胎的浓度-效应曲线,确认其LC50。方法:参照OECD推荐的方法,将受精后6 h(6 hpf)胚胎暴露在含有不同浓度马钱子的养殖水中,分别在24 hpf、48 hpf和72 hpf(96 hpf)阶段测定自主抽动次数,心率、孵化率、死亡率、畸变率等指标。结果:各暴露组24 hpf自主抽动次数、48 hpf心率以及孵化率与正常对照组都具有显著性统计学差异;48 hpf后出现脊柱弯曲、心包水肿、卵黄囊水肿,游泳异常的毒性症状;各给药组的畸形率都可达到100%;在72 hpf之后,随着浓度的升高胚胎的死亡率升高。结论:马钱子对斑马鱼胚胎发育有毒性作用。
Objective: To investigate the effect of aqueous extract of Strychnos nux - vomica on the development of zebrafish embryos and to establish the concentration - effect curve of Strychnos nux - vomica on zebrafish embryos, and to confirm its LC50. Methods: According to the method recommended by OECD, embryos 6 h after fertilization (6 hpf) were exposed to culture water containing different concentrations of Strychnos nux vomica. The numbers of spontaneous twitches were determined at 24 hpf, 48 hpf and 72 hpf (96 hpf) Heart rate, hatching rate, mortality, distortion rate and other indicators. Results: The number of 24 hpf autonomic twitches, 48 hpf heart rate and hatching rate in each exposed group were significantly different from those in normal control group. The spinal cord flexion, pericardial edema, yolk sac edema and abnormal swimming symptoms were observed after 48 hpf exposure. The rate of deformity in the treatment group was 100%. After 72 hpf, the embryo’s mortality rate increased with increasing concentration. Conclusion: Strychnos has a toxic effect on zebrafish embryo development.
