目的观察健脾解毒方对裸鼠皮下移植瘤mdr1基因及P-gp的影响。方法采用HCT8/V细胞制作裸鼠皮下移植瘤,随机分为4组,生理盐水组(空白组)、长春新碱组(化疗组)、健脾解毒方组(中药组)、长春新碱联合健脾解毒方组(联合组),观察各组多药耐药相关基因蛋白的影响。结果健脾解毒方联合化疗组mdr1基因表达水平显著低于空白组,P-gp表达量下降。结论中药复方健脾解毒方能在一定程度上可联合化疗逆转多药耐药,与化疗药具有协同作用。
Objective To observe the effect of Jianpi Jiedu Fang on mdr1 gene and P-gp in nude mice subcutaneously transplanted tumor. Methods Human xenografts in nude mice were established by HCT8 / V cells and randomly divided into 4 groups: normal saline group (blank group), vincristine group (chemotherapy group), Jianpi Jiedu Fang group (traditional Chinese medicine group), vincristine combined Jianpi Jiedu Fang group (combined group) to observe the impact of multidrug resistance-related gene protein in each group. Results The expression of mdr1 gene in Jianpi Jiedu Decoction combined with chemotherapy group was significantly lower than that in blank group, and the expression of P-gp was decreased. Conclusion Chinese medicine compound prescription of spleen and detoxification can reverse the multi-drug resistance to a certain extent and has a synergistic effect with chemotherapeutic drugs.
