以牛血清白蛋白(BSA)为酶制剂模型,用异硫氰酸荧光素(FITC)标记BSA制备了荧光蛋白质FITC-BSA。测试表明:FITC-BSA的等电点及分子质量与BSA基本相同。用FITC-BSA处理脱灰裸皮后,通过荧光显微镜能直观地观测到FITC-BSA在皮内的传递和分布情况。这些结果表明:采用本研究建立的荧光示踪技术,可以准确地检测蛋白质类材料在皮内的传质行为,该技术适用于研究制革过程酶的传质规律及作用机理。
Fluorescent protein FITC-BSA was prepared using bovine serum albumin (BSA) as an enzyme preparation model with BSA labeled with fluorescein isothiocyanate (FITC). Tests showed that the isoelectric point and molecular mass of FITC-BSA are basically the same as that of BSA. After FITC-BSA treatment, the transmission and distribution of FITC-BSA in the skin can be directly observed by fluorescence microscope. These results indicate that the fluorescence tracing technique established in this study can accurately detect the mass transfer behavior of proteinaceous materials in the skin. This technique is suitable for studying the law of mass transfer and mechanism of enzymes in tannery.
