HPLC法同时测定口溃散中盐酸小檗碱和盐酸巴马汀的含量

目的建立同时测定口溃散中盐酸小檗碱和盐酸巴马汀2种生物碱含量的方法。方法采用高效液相色谱法,色谱柱为Agilent TC-C18柱(4.6 mm×250 mm,5μm),流动相为乙腈-0.1%磷酸(47∶53,含0.1%的十二烷基硫酸钠),流速为1.0 mL/min,检测波长为268 nm,柱温为30℃。结果盐酸小檗碱和盐酸巴马汀进样量分别在0.2~1.2μg(r=0.999 9)和0.206~1.236μg(r=0.999 9)范围内线性关系良好;平均回收率分别为99.28%和97.98%(n=6),RSD分别为0.60%和1.69%。结论高效液相色谱法操作简便,准确可靠,专属性强,重复性好,可同时测定口溃散中盐酸小檗碱和盐酸巴马汀的含量,可用于口溃散的质量控制。 Objective To establish a method for simultaneous determination of two alkaloids, Berberine Hydrochloride and Palmatine Hydrochloride. Methods High performance liquid chromatography (HPLC) was performed on an Agilent TC-C18 column (4.6 mm × 250 mm, 5 μm) with acetonitrile-0.1% phosphoric acid (47:53, 0.1% sodium dodecyl sulfate) , The flow rate was 1.0 mL / min, the detection wavelength was 268 nm and the column temperature was 30 ℃. Results The calibration curves of berberine hydrochloride and palmatine hydrochloride were linear within the range of 0.2-1.2 μg (r = 0.999 9) and 0.206-1.236 μg (r = 0.999 9) respectively. The average recoveries were 99.28% and 97.98% (n = 6) with RSDs of 0.60% and 1.69%, respectively. Conclusion HPLC is simple, accurate, reliable, specific and reproducible. It can simultaneously determine the contents of berberine hydrochloride and palmatine hydrochloride in oral ulcer powder, and can be used for the quality control of oral ulcer.
