目的 了解明胶酶B在胶质细胞瘤中的作用。方法 36例胶质细胞瘤,其中Ⅰ~Ⅱ级胶质瘤13例,Ⅲ~Ⅳ级胶质瘤23例。患者术中取肿瘤组织及正常脑组织,提取组织总RNA;用逆转录共扩增定量PCR方法测定明胶酶B(MMP_9)基因表达水平。结果 Ⅲ~Ⅳ级胶质瘤明胶酶B基因表达水平(1.77±0.55)明显高于Ⅰ~Ⅱ级胶质瘤(0.63±0.11)及正常脑组织(0.46±0.08)。胶质瘤MMP_9基因表达水平与肿瘤大小无关,而与肿瘤组织分化程度有关。结论 明胶酶B在脑胶质瘤的进展中可能起重要作用。
Objective To understand the role of gelatinase B in glioma. Methods Thirty - six patients with glioma, including 13 glioma patients with grade Ⅰ ~ Ⅱ glioma and 23 glioma patients with glioma Ⅲ - Ⅳ. Tumor tissues and normal brain tissues were removed from the patients and the total RNA was extracted. The expression of gelatinase B (MMP_9) gene was determined by reverse transcription-polymerase chain reaction (RT-PCR). Results The level of gelatinase B gene in grade Ⅲ ~ Ⅳ gliomas was significantly higher than that in grade Ⅰ ~ Ⅱ gliomas (0.63 ± 0.11) and normal brain tissues (0.46 ± 0.08) (1.77 ± 0.55). Glioma MMP-9 gene expression level has nothing to do with the size of the tumor, but with the degree of tumor tissue differentiation. Conclusion Gelatinase B may play an important role in the progress of glioma.
