目的比较草乌甲素在不同方式制备的家兔破损皮肤体外渗透速率的差异,了解皮肤破损方式对草乌甲素透过率的影响。方法结合组织病理学检查,对所获破损皮肤的破损程度进行判断,同时采用改良Franz扩散池装置进行透皮试验,以草乌甲素含量为考察指标,HPLC-MS/MS法测定接受液中指标性成分含量。结果针头划伤、砂纸打磨、4%Na2S脱毛剂处理这3种方式均能得到不同损伤程度的破损皮肤;组织病理学检查显示砂纸打磨至皮肤发红时,破损程度比较适宜,局部表皮缺失,真皮完好。三者渗透速率常数存在明显差别,分别为96.82、210.2、2.821 ng/(cm·h),以砂纸打磨所获破损皮肤的渗透速率常数最大。结论砂纸打磨可以获得较好的破损皮肤模型,该模型对药物透过率影响较小。
