目的 研究E 钙黏附素 (E cad)和α 连环素 (α Cat)在胰腺癌组织中的表达与其生物学行为的关系 ,探讨胰腺癌浸润、转移的机制。方法 应用免疫组化PicTureTM二步法 ,检测 47例胰腺癌、17例正常胰腺组织 (5例正常成人胰腺和 12例远离癌组织的正常胰腺组织 )中E cad和α Cat的表达。结果 E cad和α Cat蛋白主要定位于正常胰腺腺泡和导管上皮细胞的连接处 ,17例正常胰腺组织均呈阳性表达 (+) ;5 3.2 %的胰腺癌E cad表达减低 (± ,- ) ,并与胰腺癌的组织学分级、增殖活性、淋巴结和肝转移密切相关 (分别为P <0 .0 1,0 .0 5 ,0 .0 5 ,0 .0 1) ;6 1.7%的胰腺癌α Cat表达减低 (± ,- ) ,与胰腺癌的组织学分级、淋巴结和肝转移显著相关 (分别为P <0 .0 5 ,0 .0 5 ,0 .0 1) ;E cad和α Cat表达与胰腺癌大小、侵袭能力及 1年生存率无关 (P均 >0 .0 5 ) ;同一标本中 ,E cad和α Cat的表达有显著的相关性和一致性 (r =0 .88,P <0 .0 1)。结论 胰腺癌存在E cad和α Cat基因的失活及蛋白表达下调。E cad、α Cat表达水平可作为胰腺癌增殖、分化和转移 ,尤其是肝转移的重要生物学指标。
Objective To study the relationship between the expression of E cadherin and α-catenin (α Cat) in pancreatic carcinoma and their biological behavior and to explore the mechanism of pancreatic cancer invasion and metastasis. Methods The expression of E cad and α Cat in 47 cases of pancreatic cancer, 17 cases of normal pancreas (5 cases of normal adult pancreas and 12 cases of normal pancreatic tissue far away from cancer) were detected by immunohistochemistry PicTureTM two-step method. Results E cad and α Cat proteins were mainly located in the junction of normal pancreatic acinar and ductal epithelial cells, and were positive in 17 cases of normal pancreatic tissues (+). The expression of E cad was decreased in 53.2% of pancreatic cancers (±, -) , Which was closely related to histological grade, proliferative activity, lymph node and liver metastasis of pancreatic cancer (P <0.05, 1.05, 0.05, 0.01 respectively); 1.7% The expression of cancer α Cat decreased (±, -) and was significantly associated with histological grade, lymph node and liver metastasis (P <0.05, 0.05, 0.01, respectively) The expression of Cat had no correlation with the size, invasiveness and 1-year survival rate of pancreatic cancer (all P> 0.05). There was a significant correlation and consistency between E cad and α Cat in the same specimen (r = 0.88 , P <0. 01). Conclusion The inactivation and protein expression of E cad and α Cat genes in pancreatic cancer are downregulated. E cad, α Cat expression levels can be used as an important biological indicators of proliferation, differentiation and metastasis of pancreatic cancer, especially liver metastasis.
