目的:观察银杏叶提取物(GBE)对IL-1β诱导的血管内皮细胞黏附分子表达的影响,探讨该过程中NO可能的作用机制。方法:分离培养新生牛脑微血管内皮细胞;采用ELISA方法测定E-选择素(E-selectin)、αvβ3整合素的表达。结果:GBE能够降低IL-1β诱导的E-选择素和αvβ3表达,NO增强剂L精-氨酸可进一步降低IL-1β诱导的E选-择素和αvβ3表达,而给予NO拮抗剂后GBE的抑制作用明显减弱。结论:影响NO生成可能是GBE抑制E-选择素和αvβ3整合素表达途径之一。
Objective: To observe the effect of Ginkgo biloba extract (GBE) on the expression of IL-1β-induced vascular endothelial cell adhesion molecule and to explore the possible mechanism of NO in this process. METHODS: Neonatal bovine cerebral microvascular endothelial cells were isolated and cultured. The expression of E-selectin and αvβ3 integrin was measured by ELISA. Results: GBE can reduce the expression of E-selectin and αvβ3 induced by IL-1β. The NO enhancer L-arginine can further reduce the IL-1β-induced E-selectin and αvβ3 expression, and the GBE after administration of NO antagonist. The inhibitory effect was significantly reduced. CONCLUSION: The effect of NO production may be one of the inhibitory mechanisms of GBE in inhibiting the expression of E-selectin and αvβ3 integrin.
