目的 探讨系统性红斑狼疮(SLE) 患者外周血中γδT 细胞受体(TCR)V 区基因限制性取用。方法 应用PCR特异性扩增、Southern blot 及DIG 杂交等方法检测在SLE患者和正常人外周血中TCRγ链、δ链V 区基因的表达。结果 与正常对照相比,发现SLE 患者在Vγ基因、Vδ基因均表现出限制性取用,在7 例SLE患者中均未检测到VγⅡ、Vδ4 、Vδ6 。结论 提示SLE患者TCRγδ链基因组的变化可能造成γδT 细胞的抗原识别受体表达异常而识别自身抗原,造成组织损伤。
Objective To investigate the restricted access of γδT cell receptor (TCR) V gene in peripheral blood of patients with systemic lupus erythematosus (SLE). Methods PCR-specific amplification, Southern blot and DIG hybridization were used to detect the expression of TCRγ chain and δ chain V region in peripheral blood of patients with SLE and normal controls. Results Compared with the normal control, Vγ gene and Vδ gene were found restricted in SLE patients. VγⅡ, Vδ4 and Vδ6 were not detected in all 7 SLE patients. The conclusion suggests that the change of TCRγδ chain gene in patients with SLE may cause abnormal expression of γδT antigen receptor and recognize autoantigen, resulting in tissue damage.
