复方四黄外洗液指纹图谱研究

目的建立复方四黄外洗液的HPLC指纹图谱分析方法,为该制剂的鉴别和质量标准提供依据。方法采用Zorbax SB C_(18)(150 mm×4.6mm,5μm)(美国Agilent公司));流动相为0.1%磷酸(含0.03mol/L磷酸二氢钾)(A)-乙腈(B)-甲醇(C);检测波长254nm;柱温为25℃,进样量为10μl,洗脱时间为60min,分析复方四黄外洗液的HPLC图谱,采用“中药色谱指纹图谱相似度评价系统”(2009版V1.1)进行相似度评价。结果对10批不同批号的复方四黄外洗液进行HPLC指纹图谱测定,并对其相识度进行分析,确定了13个共有峰,确认连翘苷、黄芩苷、盐酸小檗碱、大黄素峰的位置,并且各批次对照图谱间相识度高。结论建立复方四黄外洗液的HPLC指纹图谱方法简便、易行、专属性比较强,可作为复方四黄外洗液质量控制方法之一。 OBJECTIVE To establish a HPLC fingerprinting method for compound four yellow external washings and provide the basis for identification and quality standard of the preparation. Methods Zorbax SB C_ (18) (150 mm × 4.6 mm, 5 μm) (Agilent Technologies, USA) was used. The mobile phase consisted of 0.1% phosphoric acid with 0.03 mol / L potassium dihydrogen phosphate Methanol (C); detection wavelength of 254nm; column temperature of 25 ℃, the injection volume was 10μl, the elution time was 60min, the analysis of compound four Huang external wash HPLC profiles, using “Chinese medicine chromatographic fingerprint similarity evaluation system ”(2009 version V1.1) for similarity evaluation. Results Ten batches of different batch lotus root compound four Huang external washing liquid were measured by HPLC fingerprinting, and its degree of understanding was analyzed to determine the 13 common peaks, confirmed forsythin, baicalin, berberine hydrochloride, emodin peak The location, and each batch of control maps between the high degree of recognition. Conclusion The HPLC fingerprinting method of compound four yellow external washings is simple, easy and specific, and can be used as one of the quality control methods for compound four yellow external washings.
