通过将不同种类功能性油脂(亚麻籽油、油茶籽油以及菜籽油)添加到乳腺上皮细胞中,研究其对奶牛乳腺上皮细胞中SREBP基因相关信号通路表达的影响。试验选用奶牛乳腺上皮细胞(MAC-T)进行体外培养,然后在分化培养基中分别添加相同浓度的不同功能性油脂,利用实时荧光定量PCR方法检测其对甘油三酯合成基因(LPL、AGPAT3)、活化和转运基因(FASN、ACACA、SCD)表达的影响;检测其对脂肪酸网络调控基因(SREBP1C、PPARγ)表达的影响。结果表明,油茶籽油能促进ACACA、SREBP1C、FASN、AGPAT3以及SCD基因的表达,抑制PPARγ基因的表达;亚麻籽油和菜籽油对乳脂合成的相关基因基本没有促进作用,但是3种不同种类的功能性油脂均可以抑制PPARγ基因的表达。
The effects of different functional oils (flaxseed oil, camembert oil and rapeseed oil) on mammary epithelial cells were investigated to investigate the effect of SREBP gene-related signaling pathway on mammary epithelial cells. In the experiment, the mammary gland epithelial cells (MAC-T) were cultured in vitro. Different functional oils were added to the differentiation medium at the same concentration respectively. The effects of triglyceride synthetic gene (LPL, AGPAT3) , Activation and transport genes (FASN, ACACA, SCD), and their effects on the expression of fatty acid network regulatory genes (SREBP1C and PPARγ). The results showed that Camellia oleifera oil could promote the expression of ACACA, SREBP1C, FASN, AGPAT3 and SCD genes and inhibit the expression of PPARγ gene. Flaxseed oil and rapeseed oil had little effect on the related genes of milk fat synthesis, but three different kinds The functional oil can inhibit the expression of PPARγ gene.
