目的:探讨睡眠剥夺(sleep deprivation,SD)对成年大鼠海马miR132,miR138,miR127,miR128及let-7b的影响。方法:成年雄性Sprague-Dawley30只随机分为对照组(con),睡眠剥夺8 h组(sd8h),睡眠剥夺1 d组(sd1),睡眠剥夺3 d组(sd3),睡眠剥夺6 d组(sd6);采用改良多平台睡眠剥夺法(MMPM)建立大鼠SD模型,SYBRA greenⅠ荧光定量RT-PCR检测大鼠海马miRNAs的变化。结果:RT-PCR结果显示:(1)海马sd8 h组和sd1组miR132水平均低于control组(P<0.05),但sd3组高于control组(P<0.05);(2)海马sd8 h、sd3组miR138水平均低于control组(P<0.01)但sd1组高于control组(P<0.01);(3)海马sd8 h、sd1、sd3组miR127水平均低于control组(P<0.05),但sd6组高于control组(P<0.05);(4)海马sd1组miRl28水平低于control组(P<0.05),sd8h组高于control组(P<0.05);(5)海马sd8h、sdl、sd3组let-7b水平均高于control组(P<0.05)。结论:睡眠剥夺可能与海马的miR132,miR138,miR127,miR128及let-7b的水平有关。
Objective: To investigate the effects of sleep deprivation (SD) on the expression of miR132, miR138, miR127, miR128 and let-7b in adult rats. Methods: Adult male Sprague-Dawley30 rats were randomly divided into control group, sleep deprivation for 8 h group (sd8h), sleep deprivation for 1 d group (sd1), sleep deprivation for 3 d group (sd3), sleep deprivation for 6 d group sd6). SD rat models were established by modified multi-platform sleep deprivation (MMPM). SYBRA greenⅠ fluorescence quantitative RT-PCR was used to detect the changes of hippocampal miRNAs. Results: RT-PCR results showed that: (1) The level of miR132 in hippocampal sd8 h group and sd1 group was lower than that in control group (P <0.05), but higher in sd3 group than in control group (P <0.05); (2) , MiR138 in sd3 group was lower than that in control group (P <0.01), but sd1 group was higher than control group (P <0.01); (3) miR127 levels in hippocampal sd8 h, sd1 and sd3 groups were lower than those in control group (P <0.05). (4) The level of miR128 in hippocampal sd1 group was lower than that in control group (P <0.05), while in sd8h group was higher than that in control group (P <0.05) , Sdl, sd3 let-7b levels were higher than the control group (P <0.05). Conclusion: Sleep deprivation may be related to the levels of miR132, miR138, miR127, miR128 and let-7b in the hippocampus.
