目的:研究糖皮质激素骨质疏松症大鼠骨组织中PPARγ2的mRNA和蛋白表达,揭示糖皮质激素骨质疏松症的发病机理,阐述纳米钙补肾中药的调节作用机制。方法:用糖皮质激素注射的方法复制骨质疏松症模型,用纳米钙及具有益肾填精、补钙壮骨作用的补肾中药高、低剂量对实验大鼠治疗8周,以骨疏康颗粒、盖天力钙片作为阳性对照组,正常大鼠和模型空白组为空白对照组。用PCR法、Western印迹法检测骨质疏松症大鼠骨组织中PPARγ2的mRNA和蛋白表达。结果:正常大鼠骨组织可以在基因、蛋白水平表达PPARγ2,而且在骨髓脂肪细胞分化中可能起到重要的作用,糖皮质激素对大鼠骨质疏松症的发生与骨组织中PPARγ2 mRNA和蛋白表达的变化有关,从而影响骨髓脂肪细胞形成。结论:纳米钙补肾中药通过调控大鼠骨组织中PPARγ2 mRNA和蛋白表达,抑制骨髓脂肪细胞的形成,从而对糖皮质激素性骨质疏松症有一定的防治作用。
Objective: To study the mRNA and protein expression of PPARγ2 in the bone of rats with glucocorticoid osteoporosis, reveal the pathogenesis of glucocorticoid osteoporosis and elucidate the regulatory mechanism of nano-sized calcium and kidney medicine. Methods: The model of osteoporosis was established by injection of glucocorticoid. The experimental rats were treated with Nano-Calcium and Kidney-Reinforcing Kidney with high-dose and low-dose of Kidney-benefiting kidney essence and calcium supplement for 8 weeks. Granules, Capping Tianlian calcium as a positive control group, normal rats and model blank group as a blank control group. The mRNA and protein expression of PPARγ2 in bone of osteoporosis rats were detected by PCR and Western blot. Results: Normal rat bone tissue could express PPARγ2 at gene and protein levels, and may play an important role in the differentiation of bone marrow adipocytes. The effect of glucocorticoid on osteoporosis in rats was related to the expression of PPARγ2 mRNA and protein Expression changes, thus affecting the formation of bone marrow adipocytes. CONCLUSION: Nano-Calcium Bushen Chinese medicine can prevent glucocorticoid-induced osteoporosis by regulating the expression of PPARγ2 mRNA and protein in rat bone and inhibiting the formation of adipocytes.
