目的研究益气除痰方对人肺腺癌A549细胞及LEWIS肺癌小鼠肿瘤组织基质金属蛋白酶MMP1、MMP2、MMP9、MMP14等促肿瘤转移因子表达的影响。方法 :培养A549细胞,20%益气除痰方含药血清干预,划痕实验分析细胞侵袭、转移能力的改变,RT-PCR检测其对A549肺癌细胞MMP1、MMP2、MMP9、MMP14 mRNA基因表达的影响。建立C57BL/6J小鼠Lewis肺癌模型,治疗组每日予益气除痰方3.0 g.kg-1处理,灌胃21 d后,处死,检测肿瘤体积、重量、肺转移灶,并用免疫组化法比较各组肿瘤组织MMP2、MMP14的表达。结果 :20%益气除痰方含药血清可抑制A549细胞的侵袭和转移能力,并能降低MMP2、MMP14转录水平基因的表达,与空白组比较差异有统计学意义(P<0.01),MMP1、MMP9 mRNA的表达差异无统计学意义(P>0.05);益气除痰方3.0 g.kg-1组LEWIS肺癌小鼠肿瘤体积、重量、肺转移灶数明显降低,肿瘤组织MMP2、MMP14的表达降低,与模型组比较差异有统计学意义(P<0.01)。结论 :益气除痰方能抑制肺腺癌A549细胞的侵袭和转移,降低MMP2、MMP14 mRNA转录水平的表达;能抑制LEWIS肺癌小鼠肿瘤肺转移,降低其肿瘤组织MMP2、MMP14的表达,抗转移可能是益气除痰方治疗肺癌的作用机制之一。
Objective To study the effects of Yiqi Chu Tan Recipe on the expression of MMP1, MMP2, MMP9, MMP14 and other tumor metastasis factors in human lung adenocarcinoma A549 cells and LEWIS lung cancer tumor tissues. Methods: The A549 cells were cultured and the serum containing 20% Yiqi Shutan was intervened. The cell invasion and metastasis were analyzed by scratch test. The mRNA expression of MMP1, MMP2, MMP9 and MMP14 in A549 lung cancer cells was detected by RT-PCR influences. The model of Lewis lung cancer in C57BL / 6J mice was established. The treatment group was treated with 3.0 g.kg-1 of Yiqi Shutan daily, and the rats were sacrificed 21 days after the operation. The tumor volume, weight, lung metastasis were detected and detected by immunohistochemistry Method to compare the expression of MMP2 and MMP14 in tumor tissues of each group. Results: The serum of Yiqi Chu Tan Prescription could inhibit the invasion and metastasis of A549 cells and decrease the mRNA expression of MMP2 and MMP14, which was significantly different from that of the blank group (P <0.01). MMP1 (P> 0.05). The tumor volume, weight and the number of lung metastasis in LEWIS lung cancer mice of 3.0 g.kg-1 group were significantly decreased, and the expression of MMP2, MMP14 Compared with the model group, the difference was statistically significant (P <0.01). Conclusion: Yiqi Chu Tan can inhibit the invasion and metastasis of lung adenocarcinoma A549 cells, reduce the expression of MMP2 and MMP14 mRNA, inhibit the lung metastasis of lung cancer mice and decrease the expression of MMP2 and MMP14 in LEWIS lung cancer mice. Metastasis may be one of the mechanisms of Qi and Phlegm in the treatment of lung cancer.
