目的:探讨急、慢性发病过程中冠心病血瘀证心肌能量代谢的变化。方法:分别采用冠脉结扎法和异丙肾上腺素连续注射法结合高脂饲养+维生素D3复制大鼠冠心病急、慢性血瘀证模型,运用GC/TOF-MS对各模型组和对照组大鼠心肌组织进行代谢物检测,利用模式识别进行多元数据处理,筛选差异代谢物,并对差异代谢物进行相关通路分析,筛选差异显著的能量代谢通路。结果:冠心病急、慢性血瘀证各组大鼠心肌组织代谢谱与正常对照组明显不同,差异代谢物涉及能量代谢中重要的糖酵解、脂肪酸代谢和柠檬酸循环,代谢通路分析提示冠心病急、慢性血瘀证过程中能量代谢相关的差异显著通路有淀粉和蔗糖代谢、氨基糖和核苷酸糖代谢、柠檬酸循环、半乳糖代谢、不饱和脂肪酸代谢、氨基酸代谢、脂肪酸的生物合成等。结论:冠心病急、慢性血瘀证过程中心肌细胞能量代谢发生了变化,糖代谢、脂肪酸代谢、柠檬酸循环、氨基酸代谢均参与大鼠冠心病急、慢性血瘀证的过程。
Objective: To investigate the changes of myocardial energy metabolism in patients with coronary heart disease and blood stasis during acute and chronic onset. Methods: The models of acute and chronic blood stasis syndrome of coronary heart disease were established by coronary ligation and isoprenaline continuous injection combined with high-fat diet and vitamin D3 respectively. The model rats and the control group were analyzed by GC / TOF-MS The myocardial metabolites were detected by pattern recognition, and the differential metabolites were screened by pattern recognition. The differential metabolites were analyzed for related pathways, and the significant energy metabolism pathways were screened. Results: The metabolic profiles of myocardial tissue in the groups of acute and chronic blood stasis syndrome were significantly different from those in the normal control group. The differential metabolites involved in the important glycolytic, fatty acid metabolism and citric acid cycle in energy metabolism. The metabolic pathway analysis suggested that the coronary The pathways of energy metabolism in the acute and chronic blood stasis syndromes are the significant pathways of starch, sucrose metabolism, amino sugar and nucleotide sugar metabolism, citric acid cycle, galactose metabolism, unsaturated fatty acid metabolism, amino acid metabolism, fatty acid biology Synthesis and so on. CONCLUSION: The energy metabolism of cardiomyocytes changes during acute and chronic coronary heart disease. Glycometabolism, fatty acid metabolism, citric acid cycle and amino acid metabolism are all involved in the pathogenesis of acute and chronic coronary heart disease.
