潍坊地区鲜无蹼壁虎地道药材鉴别法的初步研究

目的:建立潍坊地区鲜无蹼壁虎地道药材的鉴别方法。方法:夏秋季捕捉潍坊地区成体鲜壁虎,随机抽恥100只,断食72h,水清洗晾干,95%乙醇中固定,电子天平称重,游标卡尺测长度。观察尾基部的肛疣数目特征、鼻间鳞和腹部环体一周的鳞片数目特征、指趾腹面的鳞片形状特征和指趾末端结构特征。利用十二烷基硫酸钠聚丙烯酰胺凝胶电泳技术(SDS-PACE),观察无蹼壁虎及其伪品蜥蜴的电泳迁移率,根据其蛋白电泳区带的差异。绘制电泳图谱。结果:潍坊地区无蹼壁虎头部扁平较大、呈三角形、躯干背部疣鳞极少,两侧几乎无疣鳞,大形鼻间鳞1~2枚。腹部最宽处的一横列鳞22~28枚。足指间无蹼,每指的指腹和指尖足底侧有攀瓣,一足或多足足部有暗红色斑点,分布在足背上,95%乙醇浸泡足部暗红色斑点消失。经试验鉴别,无蹼壁虎与蜥蜴的电泳图谱有显著差异,各有其特有谱带。结论:潍坊地区壁虎均为无蹼壁虎。无蹼壁虎平均体质量为(3.7±0.46)g,体长为(11.08±1.18)cm。无蹼壁虎与蜥蜴的凝胶电泳区带具有显著区别,可作为有效鉴别依据之一。 Objective: To establish the identification method of authentic medicinal materials of Gekko gecko in Weifang. Methods: The adult fresh gecko was collected in Weifang area in summer and autumn. 100 samples were taken randomly and the food was swallowed for 72 hours. After washing with water, it was air-dried, fixed in 95% ethanol, weighed with an electronic balance and measured with a vernier caliper. The number of anal warts in the tail base, the number of scales in the nasal scales and the ring in the abdomen were observed, and the shape of the scales and the structure of the ends of the digits were observed. SDS-PACE was used to observe the electrophoretic mobility of the gecko and its counterfeit lizards according to the difference of their electrophoresis bands. Draw the electropherogram. Results: Weifang region without the webbed gecko flat head large, triangular, very few warts on the back of the trunk, almost no wart scales on both sides, large nasal scales 1 to 2. The width of the abdomen at a horizontal scale 22 to 28. Fingers without webbed, finger pulp and fingertips each refers to the foot flap, one foot or more foot full of dark red spots, distributed in the dorsal foot, 95% ethanol soaked foot dark red spots disappear. The experimental identification, no webbed gecko and lizard electrophoresis patterns were significantly different, each with its own unique band. Conclusion: Weifang gecko are all non-webbed gecko. The average body mass of the gecko without webbed was (3.7 ± 0.46) g and the body length was (11.08 ± 1.18) cm. Gecko gecko and lizard gel electrophoresis zone has a significant difference, can be used as an effective basis for identification.
