以姬松茸多肽为原料,对姬松茸多肽质量指纹图谱、氨基酸组成进行分析,并对其抗氧化性及抑菌作用进行研究,为姬松茸多肽的开发提供理论依据。结果表明,所制备的姬松茸蛋白酶解产物由分子质量在850~3000Da之间的许多种多肽组成,这些多肽中8种必需氨基酸含量齐全,其中缬氨酸、赖氨酸含量较高。非必需氨基酸中谷氨酸、天门冬氨酸含量较高。姬松茸多肽具有很强的清除DPPH·、O2-·能力以及一定的抑制脂质过氧化的能力。在试验浓度范围,随着姬松茸多肽浓度的增大,清除率和抑制率逐渐增大,当其质量浓度为8mg/mL时,对DPPH·和O2-·的清除率分别达到100%、76.3%,脂质过氧化抑制率达36.4%。姬松茸多肽对金黄色葡萄球菌、大肠杆菌1515、枯草芽孢杆菌、志贺氏菌、黑曲霉、青霉等有较强的抑制作用,其抑菌率分别达72.69%、62.40%、59.88%、62.10%、62.40%、47.95%,说明该多肽中可能存在抗氧化肽和抑菌肽。
Agaricus blazei polypeptide was used as raw material to analyze the quality fingerprint of Agaricus blazei Murill polypeptide and its amino acid composition. The anti-oxidation and antibacterial activities of the polypeptide were studied, which provided a theoretical basis for the development of Agaricus blazei polypeptide. The results showed that the protease of Agaricus blazei Murill was composed of many kinds of polypeptides with molecular mass between 850 and 3000 Da. Among them, eight kinds of essential amino acids were abundant, including valine and lysine. Non-essential amino acids in glutamic acid, aspartic acid content higher. Agaricus polypeptide has a strong ability to scavenge DPPH ·, O2- · and certain lipid peroxidation. In the range of test concentration, with the increase of Agaricus blazei polysaccharide concentration, the clearance rate and inhibition rate gradually increased. When the concentration was 8mg / mL, the clearance rates of DPPH · and O2- · were 100% and 76.3 %, Lipid peroxidation inhibition rate of 36.4%. Agaricus blazei polysaccharides have a strong inhibitory effect on Staphylococcus aureus, Escherichia coli 1515, Bacillus subtilis, Shigella, Aspergillus niger and Penicillium. The antibacterial rate of Agaricus blazei Murill is 72.69%, 62.40%, 59.88% 62.10%, 62.40%, 47.95%, indicating that there may be antioxidant peptides and antimicrobial peptides in the peptide.
