目的:观察当归挥发油对左旋硝基精氨酸甲酯(L-NAME)诱导高血压大鼠血管内皮的保护作用。方法:采用灌胃L-NAME建立高血压大鼠模型,实验大鼠随机分为正常对照组、模型组、硝苯地平组及当归挥发油高、中、低剂量组,每组8只,给药组灌胃相应药物,连续8 w。比较各组大鼠收缩压变化、硝酸还原酶法检测血清一氧化氮(NO)、化学比色法检测血清一氧化氮合酶(NOS)、酶联免疫法测血清内皮素-1(ET-1)的变化,血管HE染色、扫描电镜观察血管内皮形态学特征的区别,流式细胞术计数循环内皮细胞的差异。结果:与正常对照组比较,模型组大鼠收缩压、ET-1、循环内皮细胞计数显著升高,NO、NOS显著降低(P<0.05);与模型组比较,给药组血清NO、NOS水平显著升高,收缩压、ET-1及循环内皮细胞计数显著降低(P<0.05),组织病理学结果显示模型组血管内皮受损明显,给药组较模型组明显改善。结论:当归挥发油能降低血压,改善内皮细胞损伤,对血管内皮起到一定的保护作用。
Objective: To observe the protective effect of angelica volatile oil on vascular endothelial cells induced by L-NAME in hypertensive rats. Methods: Hypertensive rat model was established by intragastric administration of L-NAME. The rats were randomly divided into normal control group, model group, nifedipine group and high, medium and low dose Angelica sinensis volatile oil groups, 8 rats in each group Group gavage the appropriate drug for 8 w. The changes of systolic blood pressure (SBP) of rats in each group were compared, the levels of serum nitric oxide (NO) were detected by nitrate reductase method, the levels of serum nitric oxide synthase (NOS) were measured by chemical colorimetry, the levels of ET- 1), vascular HE staining, scanning electron microscopy to observe the difference of morphological characteristics of vascular endothelium, and counting the difference of circulating endothelial cells by flow cytometry. Results: Compared with the normal control group, the systolic blood pressure, ET-1 and circulating endothelial cell counts in model group were significantly increased, and NO and NOS were significantly decreased (P <0.05). Compared with model group, the levels of NO, NOS The levels of ET-1 and circulating endothelial cells were significantly decreased (P <0.05). The results of histopathology showed that the vascular endothelial cells of the model group were significantly damaged and the administration group was significantly improved compared with the model group. Conclusion: Angelica Volatile Oil can lower blood pressure, improve endothelial cell injury, vascular endothelial play a protective effect.
