三七总皂苷对脑缺血再灌注损伤模型骨髓间充质干细胞动员作用的实验研究

目的探讨三七总皂苷对脑梗死后骨髓间充质干细胞动员的作用。方法参照改良ZeaLonga法建立大鼠大脑中动脉脑缺血再灌注损伤模型(MCAO)。造模成功大鼠随机分为模型组、三七总皂苷高、中、低剂量组。实验各组再随机按1、7、14、28 d时间点分为亚组。三七总皂苷高、中、低剂量组分别以60、40、20g/L浓度的血塞通胶囊粉末灌胃,每天10 mL/kg,正常组和模型组以等体积生理盐水灌胃,1次/d,共28 d。流式细胞仪检测不同时间点血液和骨髓中CD54、CD106、CD105、CD117阳性细胞百分数,ELISA法检测不同时间点血液和骨髓中干细胞因子(SCF)表达情况。结果三七总皂苷高、中剂量组和模型组的外周血和骨髓中SCF、CD117、CD105阳性细胞百分数从第1天开始升高,14 d到高峰,其后逐渐降低,但三七总皂苷高、中剂量组上升趋势优于模型组(P<0.05);模型组、三七总皂苷低、中、高剂量组外周血和骨髓中CD54、CD106在第1天到高峰,其后逐渐降低。三七总皂苷中、高剂量组与模型组相比各时间点表达均降低,其中三七总皂苷高剂量组降低明显(P<0.05)。结论三七总皂苷通过促进SCF和降低CD54、CD106表达,促进骨髓间充质干细胞从骨髓动员到外周血中,提高归巢干细胞数量。 Objective To investigate the effect of Panax notoginseng saponins on the mobilization of bone marrow mesenchymal stem cells after cerebral infarction. Methods According to the modified ZeaLonga method, a middle cerebral artery occlusion (MCAO) model was established in rats. The successful model rats were randomly divided into model group, Panax notoginseng saponins high, medium and low dose groups. The experimental groups were randomly divided into subgroups at 1,7,14,28 d points. Panax notoginseng saponins high, medium and low dose groups were 60,40,20 g / L Xuesaitong capsule powder gavage, daily 10 mL / kg, normal group and model group by equal volume of normal saline, 1 Times / d, a total of 28 d. The percentages of CD54, CD106, CD105 and CD117 positive cells in blood and bone marrow at different time points were detected by flow cytometry. The expression of SCF in blood and bone marrow was detected by ELISA at different time points. Results The percentages of SCF, CD117 and CD105 positive cells in high and middle dose group and model group increased from the first day to the peak on the 14th day, and then decreased gradually, but the total saponins of Panax notoginseng (P <0.05). The CD54 and CD106 levels in peripheral blood and bone marrow of model group and total Panax notoginseng saponins were peaked on the first day, and then decreased gradually . Panax notoginseng saponins, high-dose group compared with the model group expression decreased at all time points, of which Panax notoginseng high dose group decreased significantly (P <0.05). Conclusion Panax notoginseng saponins can promote the mobilization of SCF and the expression of CD54 and CD106, and promote the mobilization of bone marrow mesenchymal stem cells into the peripheral blood and increase the number of homing stem cells.
