目的观察抑制转化生长因子(TGF)β1表达对大肠癌细胞生长周期和侵袭力的影响。方法用小片段TGβ1的双链RNA(siRNA)转染大肠癌HCT116细胞,半定量RT-PCR法检测 TGFβ1受抑程度,流式细胞仪观察转染后细胞周期变化,软琼脂克隆实验检测肿瘤细胞生长及成瘤性的变化。结果 TGFβ1的小片段RNA转染显著抑制了肿瘤细胞TGFβ1的表达,使细胞HCT116的S 期细胞较对照组明显增高59.0%-82.5%,进入G2期细胞减少;转染组细胞克隆形成率较对照组显著下降。结论 RNA干扰技术可快捷方便地阻断TGFβ1表达,使大肠癌细胞S期阻滞并抑制肿瘤细胞生长、降低其成瘤性。
Objective To investigate the effect of inhibiting the expression of transforming growth factor β1 (TGF-β1) on the growth and invasiveness of colorectal cancer cells. Methods Human colon cancer HCT116 cells were transfected with a small fragment of TGβ1 double stranded RNA (siRNA). The degree of inhibition of TGFβ1 was detected by semi-quantitative RT-PCR. Results Small RNA transfected with TGFβ1 significantly inhibited the expression of TGFβ1 in tumor cells and increased the number of S phase cells in HCT116 cells by 59.0% -82.5% compared with that in control cells. Conclusion RNAi can block the expression of TGFβ1 rapidly and conveniently, block the S phase of colorectal cancer cells, inhibit the growth of tumor cells and reduce the tumorigenicity.
