目的探讨复方莶草合剂抗内皮细胞损伤的作用机理。方法采用高脂饲料加免疫损伤的方法,制作动脉粥样硬化内皮细胞损伤模型,用原位杂交法观察复方莶草合剂对内皮细胞一氧化氮合酶(NOS)基因表达的影响。结果高、低剂量组内皮细胞、平滑肌细胞浆内棕黄色颗粒沉积,比模型组明显增多。结论复方莶草合剂对NOS基因表达有一定的促进作用,可能是通过调节该酶、促进一氧化氮(NO)释放而起抗动脉粥样硬化内皮细胞损伤的作用。
Objective To investigate the anti-endothelial cell damage mechanism of Compound Fucao Mixture. Methods The model of atherosclerotic endothelial cell injury was established by high-fat diet and immunological injury. The effect of compound herba mixture on the expression of nitric oxide synthase (NOS) gene in endothelial cells was observed by in situ hybridization. Results The deposition of brown and yellow particles in endothelial cells and smooth muscle cells in high and low dose groups was significantly higher than that in the model group. Conclusion Compound comfrey herb has a certain role in promoting the expression of NOS gene, which may play an anti-atherosclerotic endothelial cell injury through regulating the enzyme and promoting the release of nitric oxide (NO).
