目的 建立基于原核表达的P0蛋白抗体ELISA检测方法.方法 用原核表达的P0蛋白作为包被抗原,制成固相载体.向微孔中分别加入待测样品,然后再加入辣根过氧化物酶标记抗人IgG或IgM进行反应,经过彻底洗涤后,用底物3,3’,5,5’-四甲基联苯胺(TMB)显色.TMB在过氧化物酶的催化下转化成蓝色,并在酸的作用下转化生成黄色.方法.结果 用本研究建立的P0蛋白抗体的ELISA检测方法检测正己烷低浓度接触工人190人,阳性率18.9％;检测高浓度接触工人79人,阳性率27.8％.与104名不接触正己烷健康工人(阳性率2.9％)比较,差异有统计学意义(P＜0.01).结论 基于原核表达的髓鞘P0蛋白抗体ELISA检测方法适用于大样本人群研究.
