利用DEAE-Sepharose Fast Flow离子柱层析和Sephacryl凝胶柱层析对灰树花(Grifola frondosa)子实体水提醇沉粗多糖进行分离纯化,对分离纯化到的一种多糖GFP75-2-2B进行了分析。紫外光谱鉴定结果表明,GFP75-2-2B不含蛋白质、核酸和酚类成分;离子色谱分析表明其单糖组成为岩藻糖、鼠李糖、半乳糖、葡萄糖和甘露糖,摩尔比为1.0∶1.3∶3.6∶4.0∶4.5。GFP75-2-2B对巨噬细胞株RAW264.7进行体外免疫活性的研究结果表明,GFP75-2-2B有明显刺激巨噬细胞分泌NO的活性。
The polysaccharides from Grifola frondosa fruiting bodies were separated and purified by DEAE-Sepharose Fast Flow column chromatography and Sephacryl gel column chromatography. The polysaccharide GFP75-2-2B Analyzed. The result of UV spectrum showed that GFP75-2-2B did not contain protein, nucleic acid and phenolic compounds. The analysis of ion chromatography showed that the monosaccharide composition was fucose, rhamnose, galactose, glucose and mannose, the molar ratio was 1.0 : 1.3: 3.6: 4.0: 4.5. GFP75-2-2B macrophage cell line RAW264.7 in vitro immune activity results show that, GFP75-2-2B significantly stimulate macrophage secretion of NO activity.
