目的探讨烟雾主要成分苯并芘(Ba P)对RAW264.7单核-吞噬细胞中基质金属蛋白酶(MMPs)的影响,为研究吸烟与腹主动脉瘤的关联提供相关依据。方法应用免疫印迹法观察Ba P对体外培养的RAW264.7单核-吞噬细胞基质金属蛋白酶-2/9/12(MMP-2/9/12)的影响,并验证其是否通过NF-κB信号通路。结果 Ba P暴露使培养巨噬细胞NF-κB活化,同时伴随MMP-9、MMP-12表达升高,而PDTC,核转录因子NF-κB的抑制剂,可抑制NF-κB活化,同时伴随MMP-9、MMP-12表达下降。结论 Ba P可通过NF-κB通路诱导RAW264.7单核-吞噬细胞MMP-9、MMP-12表达升高。
Objective To investigate the effects of BaP, a major component of smoke, on the expression of matrix metalloproteinases (MMPs) in RAW264.7 mononuclear phagocytes, and to provide evidence for the association between smoking and abdominal aortic aneurysm. Methods The effect of Ba P on the expression of MMP-2/9/12 and MMP-2/9/12 in RAW264.7 monocyte-macrophage phagocytes was observed by Western blotting. path. Results Ba P exposure activated macrophages NF-κB accompanied by increased MMP-9 and MMP-12 expression. Inhibitors of PDTC and NF-κB could inhibit NF-κB activation accompanied with MMP -9, MMP-12 expression decreased. Conclusion Ba P can induce the expression of MMP-9 and MMP-12 in RAW264.7 monocyte-phagocyte via NF-κB pathway.
