目的探讨胰腺癌组织中 Hedgehog 信号通路成员 GLI1 mRNA 的表达情况及其临床意义。方法采用逆转录聚合酶链反应(RT-PCR)检测25例胰腺癌组织及其相应近癌旁组织、癌旁正常组织 GLI1 mRNA 的表达,并分析胰腺癌组织中 GLI1 mRNA 表达与肿瘤大小、分化程度及转移的关系。结果 GLI1 mRNA 在胰腺癌组织、近癌旁组织及癌旁正常组织中的阳性表达率分别为68.0%(17/25)、24.0%(6/25)、0;胰腺癌组织 GLI1 mRNA 阳性表达率显著高于近癌旁组织及癌旁正常组织(均 P<0.01);GLI1 mRNA 阳性表达率与肿瘤分化程度相关(P=0.014),与肿瘤大小、有无淋巴结及远处转移无关(P>0.05)。结论 GLI1 mRNA 在胰腺癌组织中有较高的阳性表达率,且其表达率与肿瘤分化程度相关。检测 GLI1表达有助于胰腺癌的诊断,可能作为判断胰腺癌恶性程度及预后的指标。
Objective To investigate the expression of GLI1 mRNA in Hedgehog signaling pathway in pancreatic cancer and its clinical significance. Methods The expression of GLI1 mRNA in 25 cases of pancreatic cancer and its adjacent paracancerous tissues and paracancerous normal tissues was detected by RT-PCR. The expression of GLI1 mRNA in pancreatic cancer tissues was compared with that of tumor size and differentiation Degree and transfer of the relationship. Results The positive expression rate of GLI1 mRNA in pancreatic cancer tissues, paracancerous tissues and adjacent normal tissues were 68.0% (17/25) and 24.0% (6/25), respectively. The positive rates of GLI1 mRNA in pancreatic cancer tissues (P <0.01). The positive rate of GLI1 mRNA was correlated with the degree of tumor differentiation (P = 0.014), but not with the size of the tumor, lymph node metastasis or distant metastasis (P> 0.05). Conclusion GLI1 mRNA has a higher positive rate in pancreatic cancer tissues, and its expression rate is correlated with the degree of tumor differentiation. Detecting the expression of GLI1 contributes to the diagnosis of pancreatic cancer and may be used as an indicator to judge the malignant degree and prognosis of pancreatic cancer.
