目的:研究一氧化氮(NO)抑制单核内皮细胞粘附的机制。方法:以硝普钠作为NO的供体,用细胞粘附实验、流式细胞仪荧光强度测定、细胞ELISA、Northernblot等方法分别从细胞、蛋白表达、基因表达方面研究NO对内皮细胞粘附分子(vascularceladhesionmolecule,VCAM1)表达的影响。结果:NO抑制IL1α诱导的单核内皮细胞粘附,抑制IL1α诱发的VCAM1的表达并呈时间依赖性及浓度依赖性,NO在粘附分子VCAM1的基因表达水平上亦呈现抑制作用。结论:NO抑制VCAM1的表达是其抑制单核内皮细胞粘附及抗动脉粥样硬化的机制之一。
Objective: To study the mechanism of nitric oxide (NO) inhibiting monocyte-endothelial cell adhesion. METHODS: Sodium nitroprusside (NO) as NO donor was used to study the effects of NO on the expression of endothelial cell adhesion molecules (NOs) from cell, protein and gene expression using cell adhesion assay, flow cytometry, (Vascularceladhesionmolecule, VCAM 1) expression. Results: NO inhibition of IL 1α-induced monocyte endothelial cell adhesion, inhibition of IL 1α induced expression of VCAM 1 and a time-dependent and concentration-dependent, NO in the adhesion molecule VCAM 1 gene expression levels On the show also inhibitory effect. Conclusion: NO inhibits the expression of VCAM-1 is one of the mechanisms of inhibiting monocyte-endothelial cell adhesion and anti-atherosclerosis.
