目的:研究穿心莲抗菌抗病毒效价生物测定的方法,建立基于生物检测的穿心莲品质评价新模式;同时对穿心莲抗菌抗病毒效价与穿心莲内酯、脱水穿心莲内酯含量之间的相关性进行分析。方法:采用体外抑菌试验,以大肠埃希菌和金黄色葡萄球菌为菌种,采用管碟法建立生物效价评价穿心莲药材的方法,测定不同来源穿心莲药材的抗菌效价;通过血红细胞凝集试验建立穿心莲抗病毒生物效价评价穿心莲药材的方法,测定不同来源穿心莲药材抗病毒效价;同时采用HPLC法测定穿心莲中抗菌成分穿心莲内酯和脱水穿心莲内酯的含量,并运用SPSS软件分析穿心莲抗菌抗病毒效价与化学成分之间的相关性。结果:穿心莲抗菌抗病毒效价均与穿心莲内酯和脱水穿心莲内酯含量之间无显著性相关。结论:本实验所建立的生物效价检测方法可以作为穿心莲药材质量评价的方法之一。
OBJECTIVE: To study the bioassay of Andrographis paniculata and to establish a new model for the quality evaluation of Andrographis paniculata. Meanwhile, the correlation between the antibacterial and antiviral potency of Andrographis paniculata and the contents of andrographolide and dehydroandrographolide . Methods: The antimicrobial potency of Andrographis paniculata from different sources was determined by in vitro antibacterial test. Escherichia coli and Staphylococcus aureus were used as the strains. The biological potency of Andrographis paniculata was determined by tube-plate method. To establish a method for evaluating the potency of andrographis paniculata, and to determine the antiviral titer of Andrographis paniculata from different sources. At the same time, the content of andrographolide and andrographolide in Andrographis paniculata was determined by HPLC. Antibacterial and antiviral potency and chemical composition of the correlation between. RESULTS: The antibacterial and antiviral titer of Andrographis paniculata had no significant correlation with the contents of andrographolide and dehydroandrographolide. Conclusion: The biological potency test method established in this experiment can be used as one of the methods for the quality evaluation of Andrographis paniculata.
