该研究拟探索乳香没药挥发油对川芎的促透机制,观察乳香没药挥发油对角质层微观及分子结构的影响。通过测定川芎中阿魏酸在乳香没药挥发油作用下的角质层/介质分配系数,采用扫描电镜观察乳香、没药挥发油对皮肤角质层结构的影响,利用傅里叶变换红外光谱仪考察乳香、没药挥发油对角质层内角蛋白和脂质的分子结构的影响,来探索乳香没药挥发油对角质层微观及分子结构的影响。研究结果显示乳香、没药挥发油可使阿魏酸在角质层与介质分配增强,使角质层有规律的,整齐紧密堆积排列的叠瓦式结构受到一定程度的破损,同时有部分表皮鳞片向上翻起,出现局部脱落现象。另乳香、没药挥发油使角质层脂质的CH_2伸缩振动峰和角质层角蛋白的酰胺伸缩振动峰均发生了相对的位移,表明,乳香、没药挥发油可能是使角质层中脂质及角蛋白的构象发生改变,增加角质层脂质双分子层的流动性,改变有序致密结构,来达到使皮肤的通透性增加,屏障作用降低的效果。由此可知,乳香没药可能通过增加药物在角质层分配,改变皮肤角质层结构来达到促渗效果。
