目的:探讨ICAM-1在大鼠急性胰腺炎(acute pancreatitis,AP)肺损伤中的作用。方法:雄性Wistar大鼠40只,随机分为对照组(CON组)、AP不同时间点(3,6,12h)组。胆胰管逆行注射5%牛磺胆酸钠制备急性胰腺炎模型,术后3,6,12h分批剖杀大鼠,每个时间点10只。观察肺组织病理学改变并评分,检测血清淀粉酶(AMY)、肺组织髓过氧化物酶(MPO)活性;逆转录聚合酶链反应(RT-PCR)检测肺组织肿瘤坏死因子-α(TNF-α)和细胞间黏附分子-1(ICAM-1)mRNA表达;Western Blot法检测肺组织中ICAM-1蛋白表达。结果:AP组各时间点AMY、MPO和肺组织病理评分均较CON组升高(P<0.05);TNF-α和ICAM-1 mRNA表达逐渐升高,均12h达到高峰;同时ICAM-1蛋白表达也逐渐升高,并在12h达到高峰。结论:AP大鼠肺组织中,ICAM-1在mRNA和蛋白水平均呈过度表达,且与肺损伤的严重程度密切相关。前炎症因子TNF-α在肺组织中ICAM-1的过度表达起着重要作用。
Objective: To investigate the role of ICAM-1 in lung injury in rats with acute pancreatitis (AP). Methods: Forty male Wistar rats were randomly divided into control group (CON group) and AP group at different time points (3, 6, and 12 hours). Pancreatic duct retrograde injection of 5% sodium taurocholate acute pancreatitis model, after 3, 6, 12h after the rats were killed in batches, each time point 10. The lung histopathological changes were observed and scored for the detection of serum amylase (AMY) and myeloperoxidase (MPO) activity in lung tissue. The levels of tumor necrosis factor-α (TNF) in lung tissue were detected by reverse transcription polymerase chain reaction -α) and intercellular adhesion molecule-1 (ICAM-1) mRNA were detected by immunohistochemistry. The expression of ICAM-1 protein in lung tissue was detected by Western Blot. Results: The levels of AMY, MPO and pathological scores of lung tissue in AP group were higher than those in CON group at each time point (P <0.05). The expression of TNF-α and ICAM-1 mRNA increased gradually and peaked at 12h. The expression also gradually increased, reaching a peak at 12h. CONCLUSION: ICAM-1 is overexpressed in mRNA and protein levels in lungs of AP rats and is closely related to the severity of lung injury. The overexpression of pro-inflammatory cytokine TNF-α in lung tissue plays an important role.
