Using a newly developed fluorescent tracer method and illumination method, a single small molecule was observed with a light microscope using visual and photoelectric methods. Eight to 100 fluorescein isothiocyanate molecules combine to form a polyethyleneimine with a molecular weight of 20,000 and then combine into a gamma globulin. The reagent is then illuminated and probed with light of sufficient intensity to produce fully photobleached bleach during the observation. In this way, a short fluorescence pulse is generated which contains the largest possible fluorescence signal and produces a fluorescein label of about 5 to 10 x 10 5 photons / reagent molecule.
