川芎、白芷萃取物下调大鼠硬脑膜COX-2及PGE2的表达

目的:观察川芎、白芷提取物对实验性偏头痛大鼠模型COX-2及PGE2表达的影响,为都梁软胶囊治疗偏头痛提供实验依据。方法:48只健康成年Sprague-Dawley大鼠随机平均分为3组(每组16只):阳性对照组(M组)、空白对照组(CO组()生理盐水加10%DMSO,1ml/100g灌胃三天);实验组(CBM组()川芎白芷提取物,15mg/kg灌胃三天)。除空白对照组外,各组电刺激三叉神经节30min。刺激完毕取硬脑膜测定待测物质。免疫组化、蛋白质印迹(WB)法观察大鼠脑膜COX-2的表达,酶联免疫吸附法(Elisa)观察脑膜中PGE2含量的变化。结果:与CO组相比,M组、CBM组的脑膜COX-2蛋白表达、阳性颗粒数和PGE2含量显著增加(p<0.05)。与M组相比,CBM组COX-2蛋白表达、阳性颗粒数及PGE2的含量显著降低(p<0.05)。结论:川芎、白芷提取物可以抑制脑膜中COX-2、PGE2表达,推测这可能是川芎白芷控制偏头痛急性发作的重要机制之一。 Objective: To observe the effects of Chuanxiong and Angelica dahuricae extracts on the expression of COX-2 and PGE2 in experimental migraine rat model and provide the experimental evidence for the treatment of migraine by Du Luan soft capsule. Methods: Forty-eight healthy adult Sprague-Dawley rats were randomly divided into 3 groups (16 in each group): positive control group (M group), blank control group (saline group plus 10% DMSO, Gavage three days); experimental group (CBM group () Chuanxiong Angelica extract, 15mg / kg gavage three days). Except for the blank control group, each group was electrically stimulated with trigeminal ganglia for 30 min. Stimulation is completed to determine the substance to be tested dura. Immunohistochemistry and Western blotting were used to observe the expression of COX-2 in the rat brain. The contents of PGE2 in the meninges were detected by enzyme-linked immunosorbent assay (Elisa). Results: Compared with CO group, the COX-2 protein expression, the number of positive particles and the content of PGE2 in group M and group CBM increased significantly (p <0.05). Compared with M group, COX-2 protein expression, number of positive particles and PGE2 content in CBM group were significantly decreased (p <0.05). Conclusion: Rhizoma Chuanxiong and Angelica dahurica extract can inhibit the expression of COX-2 and PGE2 in the brain membrane. It is presumed that this may be one of the important mechanisms for controlling the acute attack of migraine in Angelica sinensis.
