将酸胁迫(p H=3)下的马铃薯脱毒试管苗转接于不适合试管薯诱导的MS培养基(含蔗糖30g/L)中,进行试管薯诱导,并对试管薯诱导过程中植株的蔗糖和淀粉含量及蔗糖合酶(sucrose synthase,SS)、可溶性淀粉合成酶(soluble starch synthase,SSS)和腺苷二磷酸葡萄糖焦磷酸化酶(ADP-glucose pyrophosphorylase,AGPase)的活性变化进行研究。结果表明,经酸胁迫的植株均可缩短试管薯诱导时间,经酸胁迫1d的植株在第13天开始形成试管薯,诱导时间最短,单薯重为187.8mg,单株结薯数量为2.2个,而不经酸胁迫的植株未能诱导出试管薯。植株在受酸胁迫期间,叶片中蔗糖含量无明显变化,而SS、SSS和AGPase的活性显著升高;在试管薯诱导过程中,酸胁迫植株叶片的SS活性呈降低和升高交替趋势;SSS活性在诱导试管薯0~5d时呈先下降后上升的趋势,之后趋于稳定;AGPase活性呈先上升后下降、之后再上升的趋势。
