为查明在广州罗非鱼良种场获得的第一批尼罗罗非鱼♀(Oreochromis niloticus)×萨罗罗非鱼♂(Sarotherodonmelanotheron)杂交F1群体的亲本信息,以原杂交试验中的亲本群体及杂交F1为对象,利用微卫星标记和基因重组法推测F1的亲本数目。结果发现:86对微卫星引物中有74对稳定扩增;筛选出14对在尼罗罗非鱼♀和萨罗罗非鱼♂中存在不同等位基因的引物,对杂交F1进行扩增,发现尼罗罗非鱼♀与萨罗罗非鱼♂的等位基因在F1中均结合为杂合基因型,证实杂交F1为真杂交种;应用基因型重组法根据各位点在F1中的基因型,对比其亲本群体的特异等位基因,推测F1中各位点的等位基因是来自于母本或父本,进而判断F1真实有效的父母个体的等位基因,推测出最小的有效亲本数目为一雌一雄,表明F1可能互为全同胞,为尼萨罗非鱼杂交种的进一步选育提供了基础资料。
