目的 观察单纯疱疹病毒 1型 (HSV 1)感染对体外原代培养鸡胚端脑神经元活性的影响。方法 在Vero细胞 (非洲绿猴肾细胞 )上接种HSV 1,并利用空斑实验测定病毒滴度 ;以 10 6 PFU ml病毒吸附感染原代培养鸡胚端脑神经元 ,用倒置显微镜、透射电镜、MTT细胞活性分析、DNA琼脂糖凝胶电泳 ,观察HSV 1感染对细胞活性的影响。结果 HSV 1感染神经元 2 4 ,4 8,72h与同时正常细胞比较 ,活性分别下降 2 5 % ,5 6 % ,97%。形态学及DNA电泳显示神经元主要以坏死的形式死亡。结论 HSV 1感染导致神经元活性下降。
Objective To investigate the effects of herpes simplex virus type 1 (HSV 1) infection on the activity of cultured embryonic cerebellar neurons in vitro. Methods HSV 1 was inoculated on Vero cells (African green monkey kidney cells), and the virus titer was determined by plaque assay. The primary cultured chick embryonic neurons were infected with 10 6 PFU ml of virus. The expression of HSV 1 was detected by inverted microscope, transmission electron microscopy , MTT cell activity analysis, DNA agarose gel electrophoresis, observe HSV 1 infection on cell activity. Results Compared with normal cells, the activity of HSV 1 infected neurons decreased by 25%, 56% and 97% respectively at 24, 48 and 72 hours. Morphology and DNA electrophoresis showed that neurons died mainly in the form of necrosis. Conclusions HSV 1 infection leads to the decrease of neuronal activity.
