目的 探讨雷公藤多甙 (TWH)诱导人前骨髓白血病细胞 (HL 6 0 )凋亡的作用。方法 将HL 6 0与雷公藤多甙、泼尼松共同培养 ,观察细胞的形态、细胞生长周期及DNA电泳条带的变化。结果 ⑴雷公藤多甙诱导HL 6 0细胞发生典型的细胞凋亡形态学变化 ,如细胞膜泡 (bleb)样变、细胞浆及细胞核内染色质固缩、凋亡小体出现以及DNA规律性断裂 (电泳呈典型的梯型条带变化 )。⑵细胞周期分析提示此作用首先影响增殖期细胞 ,并且呈剂量、时间依赖性。⑶雷公藤多甙诱导HL 6 0细胞凋亡的作用较泼尼松强 ;两种药物间无协同作用。结论 体外实验发现雷公藤多甙具有诱导细胞凋亡的作用 ,此可能与其免疫抑制作用以及对多种自身免疫性疾病的治疗作用及临床副作用机制相关。
Objective To investigate the effect of TWH on the apoptosis of pre-human myeloid leukemia cells (HL60). Methods HL 60 was co-cultured with tripterygium glycosides and prednisone to observe the changes of cell morphology, cell growth cycle and DNA electrophoresis bands. Results (1) Tripterygium glycosides induced typical morphological changes of HL-60 cells, such as bleb degeneration, chromatin condensation in cytoplasm and nucleus, appearance of apoptotic bodies and DNA regular rupture (Electrophoresis showed a typical ladder strip changes). (2) Cell cycle analysis suggested that this effect first affected proliferating phase cells in a dose-and time-dependent manner. ⑶ Tripterygium glycosides induced HL 60 apoptosis than prednisone stronger; no synergistic effect between the two drugs. CONCLUSION: Tripterygium glycosides can induce apoptosis in vitro, which may be related to its immunosuppressive effects and therapeutic effects on many autoimmune diseases and clinical side effects.
