目的:建立力达霉素辅基蛋白荧光衍生化的方法。方法:采用FITC和9-氨基芴分别对力达霉素辅基蛋白的氨基端和羧基端进行衍生化。偶联产物用Sephadex G15葡聚糖凝胶柱和半透膜分离纯化,并用MALDI-TOF质谱检测连接效果。结果:在本试验条件下,力达霉素辅基蛋白分别与FITC和9-氨基芴成功偶联,偶联连接分子比均为1:1。结论:此法简单和准确,可用于制备力达霉素辅基蛋白荧光衍生物。
OBJECTIVE: To establish a fluorescence derivatization method of levamycin cofactor. Methods: The amino-terminal and carboxyl-terminal of lidamycin cofactor were derivatized with FITC and 9-aminofluorene respectively. Coupling products were separated and purified on a Sephadex G15 Sephadex column and semi-permeable membrane, and the ligation effect was detected by MALDI-TOF mass spectrometry. Results: Under the conditions of this experiment, lidocaine cofactor was successfully coupled with FITC and 9-aminofluorene, respectively, and the coupling ratio was 1: 1. Conclusion: This method is simple and accurate and can be used to prepare levamycin cofactor fluorescent derivatives.
