目的观察消膏转浊方对肥胖型非酒精性脂肪肝大鼠胰岛素抵抗的影响。方法将40只Wistar大鼠随机分为5组。正常组以标准饲料喂养;其余组大鼠饲以高脂饲料。8周后,消膏转浊方组以及罗格列酮组改为普通饲料喂养,同时给予药物治疗。饮食控制组改为普通饲料喂养,共12周。于禁食次日进行钳夹实验,并计算葡萄糖输注率(GIR)。实时荧光定量PCR方法检测大鼠肝脏胰岛素受体(InsR)和胰岛素受体底物-2(IRS-2)的基因表达。结果与模型组比较,消膏转浊方组与罗格列酮组均能够显著提高肥胖型非酒精性脂肪肝大鼠的GIR(P<0.01),其InsR mRNA、IRS-2 mRNA的表达均显著高于模型组(P<0.01)。结论消膏转浊方通过增加肝脏组织InsR以及IRS-2的表达来改善胰岛素抵抗状态。
Objective To observe the effect of Xiaogan Zhuanzhuo Fang on insulin resistance in obese non-alcoholic fatty liver rats. Methods Forty Wistar rats were randomly divided into five groups. Normal group fed with standard feed; the remaining rats were fed with high fat diet. After 8 weeks, the ointment Zhunlianfang group and the rosiglitazone group were changed to ordinary feedstuffs, meanwhile medication was given. Diet control group changed to normal feed, a total of 12 weeks. Clamp test was performed on the next day after fasting, and glucose infusion rate (GIR) was calculated. Real-time fluorescence quantitative PCR was used to detect the gene expression of insulin receptor (InsR) and insulin receptor substrate-2 (IRS-2) in rat liver. Results Compared with the model group, the GXI group (all P <0.01) significantly increased the expression of InsR mRNA and IRS-2 mRNA in the obesity-non-alcoholic fatty liver rats Significantly higher than the model group (P <0.01). Conclusion Xiaotanzhunzhuo Fang improve insulin resistance by increasing the expression of InsR and IRS-2 in liver tissue.
