目的 探讨抑癌基因PTEN蛋白在人乳腺癌中的表达情况 ,及其与抑癌基因p2 7kip1 、癌基因cyclinD1表达的相关性。方法 应用免疫组织化学染色S P法 ,检测 6 1例乳腺癌组织石蜡切片中PTEN蛋白、p2 7kip1 和cyclinD1蛋白的表达情况 ,比较PTEN蛋白表达与临床病理指标的关系 ,及其与p2 7kip1 、cyclinD1蛋白表达的相关性。结果 PTEN蛋白表达定位于细胞质。PTEN蛋白阴性占6 .6 % (4 6 1) ,表达减低者占 4 1.0 % (2 5 6 1) ,阳性表达者占 5 2 .5 % (32 6 1)。PTEN与淋巴结状态、ER状态及局部复发、远处转移相关。单因素生存分析显示 ,PTEN蛋白高表达组的 5年生存率 (87.5 % )高于低表达组 (5 8.6 % ,P =0 .0 10 1)。PTEN蛋白表达与p2 7kip1 、cyclinD1表达未显示有明显相关性。结论 乳腺癌患者PTEN蛋白低表达与预后不良有关 ,PTEN对乳腺癌有一定的预后价值。在人乳腺癌中 ,p2 7kip1 和cyclinD1可能不是PTEN的主要下游基因。
Objective To investigate the expression of tumor suppressor gene PTEN in human breast cancer and its correlation with the expression of tumor suppressor gene p2 7kip1 and oncogene cyclinD1. Methods The expressions of PTEN, p27kip1 and cyclinD1 protein in paraffin sections of 61 cases of breast cancer were detected by immunohistochemical SP method. The relationship between PTEN protein expression and clinicopathological parameters was analyzed. The expressions of p27kip1, cyclinD1 Correlation of expression. Results PTEN protein was localized in the cytoplasm. The negative expression of PTEN protein accounted for 6.6% (461), while the expression of PTEN protein was 41.0% (2561), while the positive expression was 52.5% (32.61). PTEN and lymph node status, ER status and local recurrence, distant metastasis. Univariate survival analysis showed that the 5-year survival rate (87.5%) in PTEN-overexpressed group was higher than that in the low-expression group (58.6%, P = 0.010 1). PTEN protein expression and p2 7kip1, cyclinD1 expression did not show any significant correlation. Conclusion The low expression of PTEN protein in breast cancer patients is associated with poor prognosis. PTEN has a certain prognostic value in breast cancer. In human breast cancer, p2 7kip1 and cyclinD1 may not be PTEN major downstream genes.
