目的 观察青春期糖尿病大鼠生殖腺内雄激素受体的基因表达及蛋白水平变化。方法 用STZ诱导青春期大鼠糖尿病模型,实验分正常对照组、糖尿病组及胰岛素治疗组,分别以Northern blot及放射配基法检测睾丸、附睾、前列腺内雄激素受体的mRNA及蛋白水平。结果mRNA水平:生殖腺内各部位的雄激素受体mRNA表达三组间均无差异;蛋白水平:生殖腺内各部位糖尿病组的雄激素受体水平均显著低于正常组。结论 糖尿病在翻译水平干扰了雄激素受体的合成,使受体水平下降。
Objective To observe the gene expression and protein level of androgen receptor in the gonad of adolescent diabetic rats. Methods The adolescent rat model of diabetes was induced by STZ. The experimental group was divided into normal control group, diabetic group and insulin treatment group. The mRNA and protein levels of androgen receptor in testis, epididymis and prostate were detected by Northern blot and radioligand assay respectively. Results: The levels of mRNA of androgen receptor in various parts of the gonads did not differ between the three groups. Protein levels: The levels of androgen receptor in all parts of the gonads were significantly lower than those in the normal group. Conclusions Diabetes mellitus interferes with the androgen receptor synthesis at the level of translation, resulting in a decrease in receptor levels.
