目的:研究脂肪酸生物合成途径中中间代谢产物的积累情况,并推断其确切结构。方法:利用n-丁胺在牛血清蛋白(BSA)的催化下酯解酰基载体蛋白(ACP)上挂载的脂肪酸中间代谢产物,通过高分辨质谱测定得到的中间代谢产物衍生物的精确分子量,进而获得中间代谢产物的结构信息。结果:建立了一种可以有效截获脂肪酸代谢途径中间代谢物的方法,通过该方法鉴定到了13种不同的中间代谢产物,并推断了这些代谢物的结构。结论:首次确定了脂肪酸中间代谢产物以含有不饱和键的为主,部分中间代谢产物的碳链上含有羟基,导致这两类中间代谢产物的积累可能与烯脂酰ACP还原酶和3-羟基脂酰ACP还原酶的活性不足有关。
OBJECTIVE: To study the accumulation of intermediate metabolites in fatty acid biosynthesis pathway and to deduce its exact structure. Methods: The accurate molecular weight of intermediate metabolite derivatives obtained by n-butylamine catalyzed esterification of acyl carrier protein (ACP) catalyzed by bovine serum albumin (BSA) was determined by high resolution mass spectrometry. And then get the intermediate metabolite structure information. RESULTS: A method was established to intercept intermediate metabolites of the fatty acid metabolic pathway. Thirteen different intermediate metabolites were identified and the structures of these metabolites were inferred. CONCLUSION: It is the first time to confirm that the intermediate products of fatty acids contain unsaturated bonds, and some intermediate metabolites contain hydroxyl groups on the carbon chain, resulting in the accumulation of these two intermediate metabolites possibly related to the changes of acyl acyl acyl reductase and 3-hydroxy Acyl ACP reductase activity is insufficient.
