观察国产重组人白细胞介素 11(rhIL 11)对卡铂致猕猴血小板减少症的影响。成年猕猴每天静脉注射卡铂 (15mg/kg) ,连续 3天。第 3次注射卡铂后 18小时内开始皮下注射rhIL 11(每天为 5 0或 10 0 μg/kg) ,连续用药 14天。给药后定期检测外周血白细胞计数、网织血小板计数、血小板聚集功能和骨髓的集落形成和病理组织学变化等。结果发现 ,rhIL 11能明显提高注射卡铂猕猴的血小板计数最低值和缩短血小板计数少于 5 0 %药前值的持续时间 ,对白细胞和网织红细胞数的恢复也有一定程度的作用。给予rhIL 11后可见外周血网织血小板计数明显升高 ,但ADP诱导的血小板聚集功能无明显变化。rhIL 11组骨髓巨核系祖细胞和红系祖细胞集落形成增多 ,光镜下显示骨髓腔中粒系及红系细胞丰富和密集 ,巨核系细胞明显增多。上述结果表明 ,rhIL 11对卡铂致猕猴血小板减少症有明显的治疗作用 ,有助于化疗后促进多系造血功能的恢复
To observe the effect of domestic recombinant human interleukin 11 (rhIL 11) on carboplatin-induced cynomolgus monkey thrombocytopenia. Adult macaques intravenously daily carboplatin (15mg / kg) for 3 days. Subcutaneous rhIL 11 (50 or 100 μg / kg daily) was injected subcutaneously 18 hours after the third injection of carboplatin for 14 consecutive days. Peripheral blood leukocyte count, reticulocyte count, platelet aggregation and bone marrow colony formation and histopathological changes were measured regularly after administration. The results showed that rhIL 11 could significantly increase the minimum platelet count and the shortened platelet count less than 50% of the pre-drug duration in carboplatin-rhesus monkeys, which also had a certain effect on the recovery of white blood cells and reticulocytes. Peripheral blood reticulated platelet counts were significantly increased after rhIL 11 administration, but ADP-induced platelet aggregation did not change significantly. The number of rhIL11-derived myeloid and erythroid progenitor cells increased, and the myeloid and erythroid cells were abundant and dense in the marrow cavity, and the number of megakaryocytic cells increased significantly. The above results indicate that rhIL 11 has a significant therapeutic effect on carboplatin-induced cynomolgus monkey thrombocytopenia and contributes to the restoration of multi-lineage hematopoietic function after chemotherapy
